Another upgrade question- two segments

Hypothetical.

I am flying SYD to SIN, transit overnight, (<24hours), then SIN to HKG.
Can I do an online upgrade (flight booked on SQ.com) , ie, a zone 9 to zone 4 upgrade costing 35k miles (minus 15% discount), even though there are two flights involved, and on two separate days?

Yes, I am purchasing a SYD- HKG return ticket. I want to know if my one-way upgrade is going to cover both the SYD-SIN, and SIN-HKG flights.
